#### What is meant with an effect size of 0.1?

An effect size of 0.1 means that the power to detect a difference of 10% from a hypothesized null value (quantity) will be calculated. The value (quantity) under the null is determined by the fraction of negatives and by the partition volume for absolute quantification, or the expected relative quantity for relative quantities.

For example: if the relative quantity is specified as 2 and the effect size as 0.2, the resulting calculated power is the power to detect a relative quantity of 2.4.

#### Why do I need to specify the partition volume and what is the unit of the volume?

The power is affected by the between-replicate variance. For absolute quantification, the between-replicate variance is calculated on the level of the concentration, so that the partition volume affects this variance. This is also the reason why a partition volume needs to be specified for calculating the between-replicate variance.

The unit is whatever you want it to be, but make sure that the value used to calculate the between-replicate variance is the same as the one used in the power calculations. With constant partition volumes, you may safely leave the partition volume parameter at the default level. The power calculations will not be affected.
